It is also an oncoprotein that promotes cell proliferation and is upregulated in a variety of cancers, including gastric, colorectal, and non-small cell lung cancer (NSCLC) (2-4). RPRD1B contains a CTD-interacting domain (CID), which interacts with the C-terminal domain (CTD) of Pol II to regulate transcription termination (5). It is believed that this association enhances the binding of Pol II to the promoter, thereby increasing cyclin D1 transcription and promoting tumorigenesis (6). Specifically, RPRD1B promotes chromatin looping of the gene to recycle Pol II from the termination site to the promoter region (6).
