A single-pass type I transmembrane protein that is highly expressed at areas of cell-cell contact and may localize to tight junctions, MXRA8 mediates cell adhesion via interaction with Î±VÎ²3 integrin (1). Through its interactions with integrins, MXRA8 has been shown to suppress both angiogenesis (2) and osteoclast differentiation (3) by attenuating p38 mitogen-activated protein (MAP) kinase signaling. MXRA8 also plays a pivotal role in ferroptosis and the immune microenvironment of glioma, suggesting that it may serve as a novel prognostic marker and therapeutic target (4). A recent study showed that multiple emerging arthritogenic alphaviruses, including CHIKV, RRV, MAYV, and ONNV, use MXRA8 as a functional receptor (5).
