Studies show that, after feeding, increased circulating insulin and glucose activate mTORC1, which phosphorylates USP20 at Ser132 and Ser134. The phosphorylated USP20, in turn, is recruited to the HMG-CoA reductase (HMGCR) complex. HMGCR is the rate-limiting enzyme in the cholesterol biosynthesis pathway. HMGCR stabilization through deubiquitination by USP20 in the feeding state, therefore, leads to elevated cholesterol biosynthesis in the liver (2).
