PRRX1 is expressed in various types of cancer, including colon, stomach, lung, esophageal, and breast cancers. It functions as a master transcription factor of stromal fibroblasts for myofibroblastic lineage progression to promote tumorigenicity and metastasis (4). In glioma, PRRX1 promotes stemness and angiogenesis via activating the TGF-Î²\/SMAD pathway and upregulating proangiogenic factors (5). PRRX1 interacts with FoxM1 to promote the DNA Damage Response (DDR) in pancreatic cancer cells (6). The protein also drives tamoxifen therapy resistance through inducing epithelial-mesenchymal transition (EMT) in breast cancer cells (7). PRRX1 exists as two isoforms, PRRX1a and PRRX1b.
