Tetraspanins interact with a variety of cell surface proteins and intracellular signaling molecules in specialized tetraspanin-enriched microdomains (TEMs) where they mediate a range of processes, including adhesion, motility, membrane organization, and signal transduction (2,3). TSPAN8 is overexpressed in numerous epithelial cancers, where it promotes cell invasion and metastasis and is associated with poor clinical outcomes (4-6). TSPAN8 also contributes to stemness in cancer stem cells (CSCs) through activation of Î²-catenin and Sonic hedgehog (Shh) signaling pathways (7,8).
