B7-H6 is a ligand for the natural killer (NK)-cell-activating receptor NKp30, which triggers anti-tumor NK cell cytotoxicity and cytokine secretion (1). Both forms of B7-H6, cell surface B7-H6 and soluble B7-H6 (sB7-H6), are detected in primary tumors and the tumor microenvironment, but rarely in normal tissues (1-5). Cell surface B7-H6 associates with cell damage, and triggers innate immunity when it binds to NKp30 on NK cells (3,5).
