Granzymes are synthesized as zymogens and are processed into mature enzymes by cleavage of a leader sequence. They are released by exocytosis in lysosome-like granules containing perforin, a membrane pore-forming protein (2). Granzyme A, or GrA, is a dominant mediator of toxicity , and is the most abundant protease in cytotoxic granules (3). Granzyme A cleaves substrate after Arg or Lys and can activate caspase-independent cell death pathways upon cleavage of the mitochondrial protein NDUFS3.
