Studies show that mutations leading to unregulated SPT activity and increased sphingolipid synthesis cause amyotrophic lateral sclerosis (ALS), a progressive neurodegenerative disease (4). In addition, mutations in that change the substrate of SPT from serine to alanine drive the accumulation of deoxysphingolipids and cause neuropathy.
