DGK isoform zeta (DGKÎ¶) is one of the primary isoforms expressed in T cells, where it negatively regulates TCR signaling (4). DGKÎ¶ attenuates TCR activation by blocking DAG-mediated signaling of downstream effector molecules, which can result in T cell anergy (5,6). Knockout or inhibition of DGKÎ¶ has been shown to reverse T cell anergy and enhance the cytotoxicity of chimeric antigen receptor T cells (CAR-Ts), suggesting that DGKÎ¶ can be targeted for immunotherapies (7,8).
