Collectins are composed of a collagen-like region and a C-terminal carbohydrate-recognition domain (CRD) (2). During embryogenesis, COLEC11 is highly expressed in craniofacial cartilage, heart, bronchi, kidney, and vertebral bodies. It may act as a guidance cue for neural crest cell migration via recognising specific carbohydrate epitopes (3). COLEC11 is secreted in the plasma and can bind a wide range of ligands, including pathogen-associated molecular patterns (PAMPs), like lipopolysaccharide (LPS) and lipoteichoic acid (LTA) (4). After binding a PAMP, COLEC11 can activate mannan-binding lectin (MBL)-associated serine proteases (MASP1, MASP2, and MASP3) to initiate complement activation and subsequent immune processes (5,6). COLEC11 has also been shown to upregulate IL-10, but downregulate IL-6 production in retinal pigment epithelial cells (RPE) and regulate antigen-presenting cell (APC) activation in a mouse model of collagen-induced rheumatoid arthritis (7,8). Elevated COLEC11 levels in the plasma are associated with the clotting disease disseminated intravascular coagulation and often lead to organ failure (9). COLEC11 has recently been identified as a tumor growth-promoting protein by directly stimulating cancer cell proliferation and by promoting a more immunosuppressive tumor microenvironment (TME).
