ZGA occurs at the 2-cell (2C) stage of embryonic development, and ZSCAN4 is a specific marker of this stage for embryo and ES cells (2). ZSCAN4 has been shown to regulate telomere maintenance in mouse ES cells and bind to nucleosomal microsatellite DNA repeats, which are prone to genomic instability in mouse 2C-like cells (3,4). ZSCAN4 expression has also been positively correlated with derepression of heterochromatin in mouse ES cells (5).
