Both proteins have highly homologous DNA-binding domains and can synergistically regulate transcription (2,3). ERÎ² is tightly regulated by various mechanisms including estrogen binding, post-translational modifications, alternative splicing, circadian controls, and DNA methylation of the gene (4,5). The role of ERÎ² in various cancer types is controversial, but it is generally thought to be a tumor suppressor. Absence of ERÎ² has been shown to be a marker of poor prognosis in breast cancer and is correlated with tamoxifen resistance (6,7).
