Like other family members, MYO1D contains an N-terminal motor domain for actin binding, followed by two calmodulin-binding domains (IQ domains), and a C-terminal tail homology (TH1) for interaction with membrane localized proteins (1). MYO1D controls the planar cell polarity (PCP) pathway by coupling actin filament and PCP proteins to regulate cellular orientation. Using this mechanism, the protein controls cellular left-right asymmetry and direction of cilia cell movement (2-4). MYO1D also regulates myelin membrane dynamics by interacting with myelin membrane proteins (CNP and PLP) to maintain normal morphology of oligodendrocytes (5-7). The C-terminal domain of MYO1D has been shown to bind the EGFR family kinase domain (8). This interaction is required for MYO1D anchoring of EGFR family RTKs in the membrane\/cytoskeleton structure for ligand activation (9).
