The C-terminal SPR domain functions to translocate Spred1 to the membrane following growth factor stimulation (1,3). The N-terminal EVH1 domain interacts with neurofibromin to bring the later to the membrane proximal signaling complex for down regulation of Ras signaling (4,5). Mutations of Spred1 are the causes of Legius Syndrome (6,7).
