Cortisol can be found in the blood as free or bound to cortisol-binding globulins, such as transcortin. Cortisol levels follow a diurnal pattern, typically highest in the morning upon wake up and lowest just before sleep. Cortisol levels have been linked to metabolism regulation, glucogenesis promotion, liver glycogen deposition, and depression of glucose burn. In this AlphaLISA competition assay, a biotinylated cortisol tracer binds to the Streptavidin-coated Donor beads and anti-cortisol antibody associated to AlphaLISA Acceptor beads. When no cortisol is present, the beads come into close proximity. The excitation of the Donor beads triggers the release of singlet oxygen molecules, which diffuse to the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
