Reliable chemiluminescence detection In most contemporary ECL systems a luminol peroxide detection reagent is added to the membrane and reacts with the horseradish peroxidase enzyme (HRP) conjugated to the secondary antibody. HRP catalyzes the oxidation of luminol in a multistep reaction and is accompanied by the emission of low-intensity light at 428 nm, which can be measured with light-sensitive X-ray film or with a CCD imager.
