This ability is a part of Waters’ innovative approach to overcoming the challenge of retaining and separating extremely polar compounds in HILIC stationary phases. With the same selectivity as ACQUITY UPLC BEH Amide columns, the XBridge BEH Amide Column has allowed scientists to transfer separations seamlessly between HPLC and UPLC technology platforms. The XBridge BEH Amide Column is designed specifically to retain polar analytes and metabolites that are too polar to retain using reversed-phase chromatography and can support the use of a wide range of mobile phase pH, from 2 to 11. This facilitates the exceptional retention of polar analytes spanning a wide range in polarity, structural moiety, and pKa. In addition to enhancing the retention of polar compounds, the column offers an increased mass spectrometry response, direct compatibility with sample preparation eluates like PPT, LLE, and SPE, and orthogonal selectivity when compared to reversed-phase materials. The XBridge BEH Amide Column is particularly suited for the analysis of carbohydrates due to the 3.5 µm particle size, which enables high resolution and high-speed analysis in complex sample matrices while maintaining or improving the chromatographic resolution. Each batch of XBridge material is tested chromatographically with acidic, basic, and neutral analytes, with the results held to narrow specification ranges. How Should Guard Columns Be Used With The XBridge BEH Amide Column? For continued high performance of your XBridge column, use a Waters guard column of matching chemistry and particle size between the injector and the main column. It’s critical to use a high-performance and matching guard column to protect the main column without compromising or changing the analytical resolution.
Guard columns should also be replaced at regular intervals, as determined by sample contaminations. When a steady increase in system backpressure is observed above a set pressure limit, this is often an indicator that guard columns should be swapped out. Sudden appearances of split peaks can also mean this.
