This is also known as “aqueous mobile phase” or “reverse reversed-phase” because the elution order is that of a normal phase and solvents are similar to those of reversed-phase chromatography. The ACQUITY UPLC BEH HILIC Column uses Waters’ rugged 1.7 μm unbonded BEH particles in order to aid this retention and separation in very polar basic compounds. These innovative columns are optimized and tested to provide the most reproducible separations possible under UPLC HILIC conditions. ACQUITY UPLC BEH HILIC Columns are designed to overcome the major weaknesses of lab equipment that uses HILIC stationary phases, focusing on chemical stability. While silica-based HILIC phases are often chemically unstable, the rugged BEH particle offers a wide usable pH range that overcomes this instability, resulting in long column lifetimes. How Does HILIC Work? HILIC (Hydrophilic-Interaction Chromatography) is a separation technique in which a polar stationary phase is used along with a mobile phase that contains a high concentration of non-polar (organic) solvent, and a low concentration of polar (aqueous) solvent. HILIC is used primarily when compound retention can’t be achieved through traditional reversed-phase liquid chromatography.
HILIC has quickly gained popularity and a reputation as a powerful separation technique because the mobile phases used contain high concentrations of an organic solvent (such as MeCN) that are ideal for compound retention through ESI-MS.
