Product Description
AdvanceBio Sialidase A, 51 kDa. A lower mol wt version of GK80040 sialidase A for challenging digestions. Enzyme releases α(2-3)-, α(2-6)-, α(2-8), and α(2-9)-linked N-acetylneuraminic acid from oligosaccharides and glycoproteins. Also capable of releasing N-glycolylneuraminic acid. Lower mol wt sialidases may help desialylate glycans if steric hindrance is an issue. If you think a lower mol wt sialidase will help desialylate your glycans, you can also try our 88, 66 and 51 kDa Sialidase A Sampler Kit (GK80043).
Sialidases are also known as neuraminidases." Specifications: Concentration: 5 U/mL Enzyme Source: Recombinant gene from Arthrobacter ureafaciens, expressed in E. coli. Sialidases are also known as neuraminidases. Enzyme Specific Activity: ≥ 5 units/mg Enzyme Specificity: The enzyme releases a(2,3)-, a(2,6)-, a(2,8)-, and a(2,9)-linked N-acetylneuraminic acid from complex carbohydrates. The initial rate of hydrolysis of a(2,6) linkages is reported to be approximately twice that of a(2,3)-linked sialic acid however, in practice, this kinetic selectivity is of little consequence during extended incubations. Effective digestion of glycolipid substrates is facilitated by addition of a detergent, such as sodium taurodeoxycholate to the incubation. Sialidase A is capable of releasing N-glycolylneuraminic acid (Neu5Gc, NGNA) in addition to N-acetylneuraminic acid (Neu5Ac, NANA) [1], although similarly to other sialidases [2] the activity is lower toward Neu5Gc than Neu5Ac. The enzyme can also be used to remove sialic acid from gangliosides [3], glycosphingolipids (ceramide and oligosaccharide) with sialic acid. May be used for exoglycosidase sequencing with Sialidase S (GK80021), which is specific for a(2,3)-linked sialic acids. Sialidases are also known as neuraminidase. Enzyme Unit Definition: One unit is defined as the amount of enzyme required to catalyze the release of 1 µmole of p-nitrophenol from p-nitrophenyl-α-D-N-acetylneuraminic acid per minute at 37° C, pH 5.5. Molecular Weight: 51 kDa Volume: 200 µL pH Optimum: 6 **********The following product description is generated by AI and is for reference only. If you have any questions, please contact customer service.********** AdvanceBio Sialidase A, 51 kDa, is a highly purified enzyme specifically designed for efficient and reliable cleavage of terminal sialic acid residues from glycoproteins and other glycoconjugates. This reagent enables precise structural analysis of glycan compositions by removing sialic acid groups, thereby facilitating downstream applications such as glycan profiling, characterization of biotherapeutics, and assessment of glycosylation patterns in protein research and development. Ideal for both academic and pharmaceutical laboratories, this sialidase is well-suited for use in workflows involving released glycans and intact glycoprotein analysis. Compatible with a range of analytical platforms, including liquid chromatography and mass spectrometry, it integrates seamlessly with Agilent’s AdvanceBio Glycan Mapping columns, glycoprotein sample prep kits, and related biomolecule characterization systems. By providing robust and consistent activity under recommended conditions, this enzyme supports method development and routine analysis for biopharmaceutical quality control, comparability studies, and biosimilar development. Each lot undergoes stringent quality control to ensure batch-to-batch consistency and reproducible results, making this product a valuable tool for enhancing accuracy in glycan analysis and bioprocess monitoring.
