Product Description
Xcellerex APS main instrument The Xcellerex™ Automated Perfusion System (APS) is designed to streamline upstream perfusion applications by integrating multiple functions into a single-use system. Here are some key benefits of the system:
• Simplify operations: Integrate all perfusion operations into a single platform, including bioreactor, cell retention, fluid management, and automation. Access everything via one user interface that generates a single batch report. • Minimize capital expenditure: Use a single system that supports your volume needs from less than 50 L to 500 L. • Achieve high-density cell cultures: The low-shear recirculation pump minimizes impact on cell viability, ensuring high-density cultures. • Maintain optimal filter performance: Intelligent filter switching minimizes user intervention and eliminates the need for a backup system. • Reduce manual interactions: Automated liquid management controls media and permeate bin switching, reducing the need for manual intervention. • Maximize flexibility: Utilize standard ReadyToProcess™ hollow fiber cartridges in a wide range of sizes for both microfiltration and ultrafiltration applications. See the system in 3D | View data file (PDF) Key Xcellerex™ ASP system components and features: Pumps Recirculation pump: Type: Levitronix™ single-use maglev centrifugal pump. Properties: Low-shear, ideal for cell culture perfusion applications. Flow Rate: Operates between 1 to 18 liters per minute (LPM). Peristaltic pumps: Functions: Variable speeds for media addition, cell bleed, and permeate delivery. Applications: Media Addition: Adds fresh media to the bioreactor. Cell Bleed: Maintains steady state by removing excess cells. Permeate Delivery: Transfers permeate to collection bins. Filter holder Cartridge Type: Operates with a maximum of two ReadyToProcess™ hollow fiber cartridges. Orientation: Mounted vertically with feed inlet at the bottom and retentate outlet at the top. Flexibility: Supports multiple cartridge and pore sizes for microfiltration (MF) and ultrafiltration (UF) applications. Xcellerex™ APS flow kit Presterilized: Gamma irradiated, bioburden free, ready for use. Sections: Four sections for recirculation feed, permeate, retentate, and perfusion media. Components: Includes single-use pump head, tubing, pre-calibrated sensors for pressure and flow, ReadyMate™ connectors. Installation: Color schemes for ease of installation. Recirculation flow path Components: Recirculation feed line, ReadyToProcess™ hollow fiber filter, retentate line. Function: Transfers cell culture supernatant from bioreactor to filter. Cell Harvest: Allows bioreactor to be emptied post-run without external pump. Filter Management: Supports up to two filters with intelligent switch feature based on performance. Permeate flow path Function: Directs product or spent media to collection vessel. Automated Switch: Automatically switches to new collection vessel when full. Components: Supports up to two single-use permeate collection bags, pressure sensor, flow meter, sampling line. Perfusion media flow path Function: Delivers perfusion feed media to bioreactor. Components: Supports up to two single-use media storage bags on weigh scales for continuous weight monitoring. Cell bleed flow path Function: Achieves desired cell density through automated cell bleed. Components: Bin on weigh scale for feedback, supports continuous or bolus bleed mode. Perfusion bag Design: Seamless connectivity to Xcellerex™ bioreactor and flow kits, designed for perfusion applications. Alternating permeate flow (APF) Function: Defines high and low setpoints for permeate flow to encourage filter longevity by preventing lodging of cell debris. Integrated control with AVEVA™ software Software: AVEVA™ (formerly Wonderware™) running on Microsoft® Windows® server. User Interface: X-Station with touchscreen, keyboard, and mouse. Features: Perfusion overview screen, status of instrument modules, alarm and event notifications, graphical user interface (GUI) for monitoring and control.
