Product details:
This antibody is useful as a general reagent for the detection of axonal growth cones in tissue sections and potentially a key marker in neuronal development research.
This antibody recognises an intracellular protein 2G13p which is restricted to growth cones in developing brain and embryonic neurons in culture. The protein 2G13p also appears to be associated with filamentous actin in the growth cones suggesting a role for 2G13p in growth cone motility. This antibody is suitable for use in immunohistological studies of both frozen and paraffin-embedded sections, staining growth cones, including their filopodia and lamellipodies.

Supplementary Information:
This supplementary information is collated from multiple sources and compiled automatically.
The growth cone also known as the neuronal growth cone is a dynamic structure found at the tip of a growing axon or dendrite in neurons. It weighs approximately 1.2 million Daltons and is comprised of a complex network of proteins and membrane-bound components. Growth cones are primarily expressed in the developing nervous system where they play an important role in guiding axons and dendrites to their synaptic targets. Essential molecules include actin and tubulin which provide structural support and are involved in movement through polymerization and depolymerization.
Biological function summary
Growth cones are critical for neuronal development and network formation. They function as sensory-motile organelles that explore the extracellular environment responding to guidance cues such as soluble gradients or substrate-bound cues. They translate these signals into directed movement and steering decisions for axonal pathways. Growth cones operate not just as individual units but are part of larger cellular complexes coordinating with other cytoskeletal structures and signaling molecules such as calcium ions and cyclic nucleotides to modify extension and retraction dynamics.
Pathways
Growth cones engage in key mechanisms like the Wnt signaling and PI3K/AKT pathways. These pathways modulate cytoskeleton remodeling and intracellular calcium levels which in turn influence growth cone motility and directionality. Within these pathways growth cones interact with proteins such as β-catenin in Wnt signaling providing cross-talk regulation necessary for synapse formation and neuronal plasticity.
Growth cone malfunction is associated with conditions such as spinal cord injuries and neurodegenerative diseases like Alzheimer's. Aberrant signaling within growth cones often involving proteins like amyloid precursor protein (APP) leads to impaired axonal guidance and regeneration. Understanding growth cone behavior and its association with pathology offers insights into potential therapeutic strategies for promoting neural repair and mitigating neurodegenerative progress.
