Abcam's Human Endostatin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of Human Endostatin in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for Human Endostatin coated on a 96-well plate. Standards and samples are pipetted into the wells and Endostatin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Endostatin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Endostatin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Endostatin also known as Collagen XVIII alpha 1 (COL18A1) is a powerful anti-angiogenic protein with a mass around 20 kDa. It specifically inhibits the process of blood vessel formation in both in vitro and in vivo environments. This protein derives as a fragment from the carboxyl end of collagen XVIII which is a component of basement membranes. Expression of endostatin occurs primarily in the endothelial cells lining blood vessels and is also seen in tissues undergoing remodeling or repair.
Biological function summary
Endostatin functions as an important inhibitor of angiogenesis the process in which new blood vessels form from pre-existing ones. It acts independently and is not part of a larger protein complex. In the cellular microenvironment endostatin binds to various integrins inhibiting endothelial cell proliferation and inducing apoptosis. It also disrupts signal transduction pathways critical for endothelial cell survival and migration.
Pathways
Endostatin holds an important position in the angiogenesis and Wnt signaling pathways. It interacts closely with proteins like VEGF (Vascular Endothelial Growth Factor) and FGF2 (Fibroblast Growth Factor 2) where it counteracts their pro-angiogenic effects. Endostatin's role in these pathways indicates its significant impact on controlling angiogenic balance in various physiological and pathological conditions.
Endostatin exhibits notable relevance to cancer and diabetic retinopathy. In cancer the protein suppresses tumor-induced angiogenesis restricting tumor growth and metastasis. Likewise in diabetic retinopathy endostatin's inhibition of abnormal blood vessel formation in the retina offers potential therapeutic benefits. In these contexts it interacts with other key proteins like MMPs (matrix metalloproteinases) which regulate collagen degradation and release of endostatin fragments from parent collagen XVIII.
