ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgG class antibodies against Epstein Barr virus nuclear antigen (EBNA) in Human serum and plasma.
A 96-well plate has been precoated with Epstein Barr virus antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Epstein Barr virus-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Epstein Barr virus IgG sample captured in plate.

Epstein-Barr virus nuclear antigen (EBNA) IgG is an antibody that targets the Epstein-Barr virus (EBV) which is known for its role in infectious mononucleosis. EBNA proteins are expressed in the lymphocytes infected by EBV primarily B-cells. They play a mechanical role in viral latency and promote cell survival and proliferation. EBNA-1 the most studied of the EBNA family is a protein with a mass of approximately 72 kDa. It is essential for maintaining the episomal form of the EBV genome within the host cell.
Biological function summary
EBNA proteins modulate host cell functions linking them to cell cycle regulation and transcriptional control. They contribute to the transformation of B-cells by interacting with components of the host's cellular machinery. EBNA is not known to form part of a larger protein complex but interacts with host proteins such as the retinoblastoma protein (Rb) and p53 impacting their pathways.

EBNA IgG is intricately involved in the EBV latency program and the associated signaling pathways that control cell proliferation and survival. It is part of the pathways that include the NF-kB signaling and apoptotic regulation pathways. EBNA interacts with proteins like LMP1 an EBV-encoded protein that activates NF-kB signaling further integrating itself into the complex network of host-pathogen interactions.
EBNA IgG is significantly associated with the pathogenesis of infectious mononucleosis and certain autoimmune conditions such as multiple sclerosis. In these disorders EBNA interacts with host proteins like Rb leading to cell cycle dysregulation and immune system challenges. The presence of anti-EBNA IgG plays an important role in diagnosing past infection with EBV and assessing the risk for EBV-associated diseases.
