PEP12 also known as Vps45-dependent receptor or Vps45 is a protein that plays a mechanical role in vesicle trafficking. It is involved in the fusion of vesicles with their target membranes participating in the transport of proteins and lipids within cells. The protein with a molecular mass of approximately 12 kDa is expressed in yeast and other eukaryotic organisms. It primarily localizes to membranes of the Golgi apparatus and vacuoles where it participates in membrane fusion events necessary for intracellular transport.
Biological function summary
PEP12 contributes to the selective docking and fusion of vesicles. As a part of the SNARE complex an important structure for vesicle fusion PEP12 aids in tethering docking and the eventual fusion of vesicles and their target membranes. It works alongside proteins such as Vps45 and other SNARE components coordinating vesicular trafficking in cellular compartments. This role ensures efficient sorting and proper distribution of lipids and proteins essential for cell function and homeostasis.
Pathways
PEP12 functions in the secretory and endocytic pathways. In the secretory pathway it ensures the delivery of cargo from the trans-Golgi network to the vacuole or lysosome. In the endocytic pathway PEP12 is involved in recycling molecules back to the Golgi or in their delivery to other cellular destinations. The protein interacts closely with other SNARE proteins and factors like Sec1p family members which regulate its activity and ensure precise targeting of vesicles during these processes.
PEP12's role in vesicle trafficking links it to various conditions characterized by faulty protein and lipid transport. Research has indicated a connection between defects in components of the SNARE complex including PEP12 and neurodegenerative disorders such as Alzheimer's disease. The protein also interfaces with Rab proteins which are central to vesicle budding movement and fusion. Errors in vesicular trafficking could result in accumulation of cellular waste contributing to disease pathogenesis highlighting the need to understand PEP12's functions for potential therapeutic targeting.
