Cingulin also known as CJ and 140 kDa protein is an integral component of the tight junction complex. It predominantly resides in the cytoplasm near cell membranes specifically in epithelial and endothelial cells. The primary mechanical role of cingulin is to link the actin cytoskeleton to tight junction proteins thereby facilitating cellular barrier functions. Cingulin works as a scaffold to anchor other junctional proteins and helps maintain the architecture of epithelial cellular sheets.
Biological function summary
Cingulin functions by controlling paracellular permeability ensuring that substances pass through cells in a regulated manner rather than between them. Cingulin is part of the larger tight junction complex and associates with other proteins like ZO-1 and occludins. The protein modulates signaling pathways that dictate cell proliferation and differentiation which are key processes in maintaining tissue integrity and enabling proper cellular communication.
Pathways
The interaction of cingulin is notably important in the Wnt signaling pathway and the RhoA pathway. These pathways contribute to cellular activities such as proliferation and migration. Cingulin's interaction with protein kinase C and GEF-H1 links it to the regulation of cytoskeletal dynamics and cell cycle progression. Its role within these pathways highlights its importance in maintaining tight junction stability and modulating intracellular signaling processes.
Cingulin has significant connections to inflammatory bowel disease and certain cancers. Dysregulation of cingulin expression and function are linked to altered barrier permeability and disrupted cellular organization contributing to pathogenesis. In these disorders alterations in related proteins such as claudins and junctional adhesion molecules are often observed complicating the cellular landscape and affecting tissue health. Cingulin's role in disease emphasizes its importance in barrier maintenance and cellular signaling.
