Elastase also known as CEL-A3B refers to a family of serine proteases with an approximate mass of 29 kDa. It primarily resides in the pancreas and neutrophils where it performs key enzymatic roles by breaking down elastin and other proteins. Humans express elastase in various forms including pancreatic elastase and neutrophil elastase. In these tissues elastase also participates in immune defense and proteolysis of extracellular matrix components.
Biological function summary
Elastase serves multiple functions beyond simple protein degradation. As a component in various complexes such as those found in neutrophils elastase helps in modulating inflammatory responses. It cleaves secreted factors and membrane proteins affecting cell signaling and recruiting additional immune cells. This protease also regulates tissue remodeling by degrading unwanted matrix proteins facilitating cellular migration and healing processes.
Pathways
Elastase significantly impacts inflammatory and tissue repair pathways. It interacts with proteins such as alpha-1 antitrypsin which serves as a natural inhibitor helping to control elastase activity. In particular elastase plays a role in the inflammatory response pathway breaking down cytokines and chemokines and is also implicated in fibronectin degradation part of the tissue repair process. Through these interactions elastase helps to modulate the balance between proteolysis and tissue preservation.
Elastase exhibits a notable connection to conditions like chronic obstructive pulmonary disease (COPD) and cystic fibrosis. Overactivity or misregulation of elastase can lead to destructure of lung tissue contributing to COPD. In cystic fibrosis mucus buildup leads to heightened inflammation and tissue damage partly due to unregulated elastase activity. Both diseases show interactions with proteins like alpha-1 antitrypsin where deficiencies further exacerbate elastase's pathological effects highlighting its clinical importance.
