The Newcastle disease virus Hemagglutinin-neuraminidase commonly referred to as HN is a multifunctional protein expressed on the surface of the Newcastle disease virus (NDV). This protein assists in viral attachment and spread by displaying both hemagglutination and neuraminidase activities. With a molecular mass of approximately 74 kDa HN binds sialic acid receptors on host cell surfaces to facilitate viral entry. It is expressed on the viral envelope and plays a role in promoting fusion with host cell membranes by interacting with the NDV fusion (F) protein.
Biological function summary
HN supports the infectivity of the Newcastle disease virus by binding to and cleaving sialic acid residues on glycoproteins across species. This cleavage releases newly formed viral particles from host cells enabling further spread within the host. It is an important element of the NDV as it forms transient complexes with other viral proteins such as the F protein. This interaction enhances the HN’s ability to mediate membrane fusion which is essential for the establishment of infection.
Pathways
The Newcastle disease virus Hemagglutinin-neuraminidase integrates into the viral replication pathway by influencing host-virus interactions and facilitating membrane fusion. It participates in the viral life cycle alongside pathways involving its close relative the F protein which contributes to the fusion of viral and host cell membranes. The interplay between HN and F protein in initiating and propagating infection highlights the protein's role in viral pathogenicity.
Newcastle disease virus Hemagglutinin-neuraminidase directly connects to Newcastle disease a highly contagious avian disease leading to significant economic impact in poultry farming. HN associates with the immune evasion strategies of NDV often leading to neurological and respiratory symptoms. In its viral context HN not only interacts with the F protein but also contributes to host immune system evasion perpetuating disease symptoms and complicating therapeutic intervention strategies.
