CBP80 also known as cap-binding protein 80 is an important component of the cap-binding complex (CBC) with a mass of about 80 kDa. This protein primarily binds to the 5' cap structure of newly synthesized pre-mRNA in eukaryotic cells. CBP80 often associates with CBP20 and together they form the CBC. It facilitates the processing of pre-mRNA and is mainly expressed in the nucleus of cells. CBP80 is integral in stabilizing RNA transcripts during their maturation.
Biological function summary
CBP80 plays a role in the initiation of mRNA processing events. As a part of the CBC it influences RNA splicing export and translation initiation. It participates in early mRNA export from the nucleus to the cytoplasm and is essential in quality control mechanisms that ensure accurate mRNA translation. The CBP80-CBP20 complex also impacts nonsense-mediated mRNA decay (NMD) a process that degrades faulty mRNAs preventing the production of truncated proteins.
Pathways
CBP80 integrates into the mRNA surveillance pathway where it works closely with other key proteins such as UPF1 UPF2 and UPF3 in the NMD pathway. These interactions ensure erroneous mRNA is efficiently targeted and degraded. Additionally CBP80 plays a minor role in the pathway of general mRNA export where it cooperates with export factors like CRM1. Its involvement in these pathways highlights its role in maintaining the fidelity of gene expression.
CBP80's malfunction has been linked to certain cancer types and neurodevelopmental disorders. Altered activity of CBP80 can lead to the accumulation of aberrant mRNA transcripts which contributes to oncogenesis. In some neurodevelopmental disorders mutations affecting CBP80 or its interacting partner CBP20 can disrupt normal neural development. This suggests that CBP80 not only maintains normal cell function but also prevents the development of disease through its regulation of gene expression.
