After synthesis in magnocellular neurons of the supra-optic and paraventricular nuclei, the precursor is packaged into neurosecretory vesicles and transported down the axon to the posterior pituitary (neurohypophysis). During this axonal transport the precursor is proteolytically cleaved, generating mature oxytocin and its tightly bound neurophysin I; the complex is stored in nerve terminals and secreted into the systemic circulation upon appropriate stimulation.
