RhoF also known as Rif or Rho-related GTP-binding protein RhoF is a member of the Rho family of small GTPases. It has a molecular mass of around 21 kDa. RhoF acts as a molecular switch cycling between an active GTP-bound state and an inactive GDP-bound state. It is known to regulate the organization of the actin cytoskeleton. RhoF expression is quite marked in tissues like the brain and testis where it carries out its functions by interacting with various effector proteins.
Biological function summary
RhoF plays a role in organizing the actin cytoskeleton and is not found as part of a larger protein complex. It triggers the formation of filopodia and other actin-associating structures influencing cell shape and movement. RhoF is central to processes that depend on dynamic actin remodeling such as cell migration adhesion and morphogenesis. Through these interactions it impacts cell functionality in various cellular contexts.
Pathways
Its activity is associated with the regulation of actin cytoskeleton pathways. RhoF interacts with proteins like Cdc42 and Rac1 members of the same Rho GTPase family which also participate in these pathways. It contributes to the signaling networks that govern cytoskeletal rearrangements and influences cellular dynamics. This integration into these pathways allows RhoF to influence downstream effector pathways assisting cells in responding to extracellular cues.
RhoF's role in actin cytoskeleton regulation links it to conditions involving cytoskeletal dysfunction such as certain types of cancers and developmental disorders. In cancer its interaction with related proteins like Rac1 may influence tumor cell migration and metastasis. In developmental disorders improper regulation of RhoF and its signaling pathways can result in abnormal cell morphology and motility affecting organ development and function. These associations make RhoF a potential target for therapeutic interventions in these disorders.
