SAT1 also known as Spermidine/Spermine N1-acetyltransferase 1 facilitates the acetylation of polyamines such as spermidine and spermine. It has a molecular mass of approximately 21 kDa. SAT1 primarily functions in the cytoplasm and has significant expression in tissues like liver kidney and brain. As an enzyme SAT1 catalyzes the transfer of acetyl groups to its polyamine substrates an important step that regulates polyamine levels within cells.
Biological function summary
Regulation of polyamine concentrations is important for cellular growth differentiation and apoptosis. SAT1 acts as a gatekeeper for polyamine levels by controlling their acetylation marking them for degradation or export out of the cell. The enzyme does not form part of a larger complex but operates independently to modulate polyamine homeostasis. By converting excess polyamines into acetyl derivatives SAT1 maintains cellular balance and ensures normal cellular functions.
Pathways
SAT1 plays important roles in the polyamine metabolic pathway and the urea cycle. Within the polyamine pathway SAT1 interacts closely with proteins like Spermine Synthase (SMS) and Spermidine Synthase (SRM). These proteins contribute to a tightly regulated process where SAT1's acetylation activity facilitates the recycling and eventual degradation or export of polyamines. This regulation aligns with the need for cell cycle progression and normal cellular physiology.
SAT1 shows links to conditions such as cancer and neurodegenerative diseases. Dysregulation of polyamine metabolism where SAT1 plays a central role connects to cancer development due to uncontrolled cell proliferation. Additionally neurodegenerative diseases like Alzheimer's disease also exhibit alterations in polyamine levels with changes in SAT1 activity influencing disease progression. In these contexts related proteins like Antizyme 1 (OAZ1) work alongside SAT1 in modulating polyamine homeostasis showing the broader biological impact of this enzyme.
