ZBTB4 also known as Zinc Finger and BTB Domain-Containing Protein 4 is a transcriptional repressor with a mass of approximately 140 kDa. ZBTB4 contains a BTB/POZ domain and several zinc finger motifs allowing it to bind DNA and suppress gene expression. It is expressed in various tissues with higher levels observed in the brain lung and liver. The protein primarily localizes in the nucleus where it regulates the transcription of specific target genes.
Biological function summary
ZBTB4 interacts with other proteins as a part of transcriptional repression complexes. Its role includes modulating gene expression linked to cellular proliferation and differentiation. ZBTB4 often works together with corepressors such as CtBP which enhances its repressive function. Through these interactions it helps maintain genomic stability and manage cell cycle progression.
Pathways
Research indicates that ZBTB4 is an important component of the p53 signaling and apoptosis pathways. It plays a role in mediating the expression of genes involved in cell cycle arrest and apoptosis which are critical in response to DNA damage. The p53 tumor suppressor protein a well-known pathway partner interacts with ZBTB4 to control cell fate decisions under stress conditions.
ZBTB4 is implicated in cancer progression and development. Alterations in ZBTB4 expression or function can contribute to tumorigenesis often relating to its connections with p53 and MDM2 proteins. This linkage points to a role in breast cancer and gastric cancer where deregulation of cell cycle and apoptosis pathways is common. Understanding the involvement of ZBTB4 in these pathways could offer insights into new therapeutic strategies.
