Pbx3 also known as pre-B-cell leukemia transcription factor 3 is a transcription factor that belongs to the TALE/PBX homeobox family. It has an approximate mass of 47 kDa and usually expresses in tissues such as the brain and the pancreas. Mechanically Pbx3 regulates gene expression by binding to specific DNA sequences and forming complexes with other transcription factors. Its function is important for the transcriptional regulation process during developmental stages.
Biological function summary
Pbx3 plays a significant role in the development of several tissues including the central nervous system. It frequently associates with other proteins like Meis1 and HoxB1 to form transcriptional complexes that regulate essential genes involved in cell differentiation and organogenesis. These complexes mediate the control of target gene activity ensuring proper embryonic development and cellular differentiation.
Pathways
Pbx3 participates in critical regulatory networks such as the retinoic acid signaling pathway and the hematopoietic stem cell differentiation pathway. In these pathways Pbx3 acts in conjunction with proteins like Hox genes and Meis proteins influencing the transcriptional regulation important for tissue specification and stem cell renewal. This functional interaction allows Pbx3 to contribute significantly to developmental processes and cellular programming.
Pbx3 has significant implications in conditions like acute lymphoblastic leukemia and diabetes mellitus. In acute lymphoblastic leukemia Pbx3 interacts with oncogenic fusion proteins such as E2A-Pbx1 contributing to the malignancy's development by altering normal transcriptional regulation. In diabetes its expression levels affect pancreatic development and insulin regulation linking it to metabolic disturbances associated with the disorder.
