Hepatitis C Virus E1 (HCV E1) also known as glycoprotein E1 is a component of the viral envelope with a mass of approximately 31 kDa. It plays a role in viral entry into host cells by binding to cellular receptors and facilitating fusion with the host membrane. E1 is expressed on the surface of Hepatitis C virions providing structural support. It forms a heterodimer with another envelope protein E2 which is essential for its functionality in the virus life cycle.
Biological function summary
The Hepatitis C Virus E1 protein engages in the process of infecting liver cells. As part of the heterodimeric E1/E2 complex it enables the virus to attach and penetrate host cells which is important for establishing infection. The E1/E2 complex is also important for immune evasion as it can escape neutralizing antibodies due to its high mutation rate. This complex allows Hepatitis C to persist in the host leading to chronic infection.
Pathways
E1 and its partner E2 are integral to the viral replication cycle. The proteins interact with host cell pathways that dictate cell entry and immune response. E1 involves itself in the fusion process and is linked to the endocytosis pathway. Other proteins related through this pathway include CD81 a known receptor for HCV which facilitates the actual uptake of the virus into the host cell.
Hepatitis C Virus E1 plays a significant role in chronic hepatitis C infection and hepatocellular carcinoma. This infection leads to liver inflammation which can progress to fibrosis or even liver cancer. E1 also interacts with proteins like claudin-1 which supports tight junction formation in liver cells and contributes to the spread of the virus enhancing the risk of disease progression. Understanding the function of E1 is important for developing therapies and vaccines against HCV.
