Abcam's Furin Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of Human Furin in serum, plasma (collect plasma using heparin and EDTA as an anticoagulant. Citrate is not recommended) and cell culture supernatants.
This assay employs an antibody specific for Human Furin coated on a 96-well plate. Standards and samples are pipetted into the wells and Furin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Furin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Furin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimization may be required with urine samples.

Furin is a serine endoprotease that plays critical role in the cleavage of precursor proteins into their active forms. It belongs to the proprotein convertase family and is also known as furin protease. The molecular weight of furin is approximately 54 kDa. Furin is ubiquitously expressed in various tissues in the body with high expression levels in the liver and the central nervous system. Furin is mainly located in the trans-Golgi network where it processes many precursor proteins involved in various cellular functions.
