C5 also known as the C5 protein is an essential component of the complement system. It is a large glycoprotein with a molecular weight of approximately 196 kDa. It circulates in the blood plasma as part of the immune system where it remains inactive until cleaved into two fragments C5a and C5b. The fragment C5a or C5-a acts as a potent anaphylatoxin and is an important mediator in inflammatory responses. C5 is ubiquitously expressed in various tissues but is abundantly found in liver tissue.
Biological function summary
The role of C5 and its cleavage products are significant in the immune response. C5a a powerful chemoattractant promotes recruitment of neutrophils and other inflammatory cells at sites of infection. C5b initiates the formation of the membrane attack complex (MAC) which is important for lysing pathogens. C5a as part of the C5/C5a axis plays a vital role in regulating immune cell activity and modulating inflammatory processes. The protein is part of the terminal complement complex engaging in immune defense mechanisms.
Pathways
C5 is integral to the classical and alternative complement pathways. In both pathways the cleavage of C5 is a critical step that leads to the eventual formation of the MAC. C5 interacts with several proteins including C3 and C6 to execute its functions effectively. The close relationship between C3 activation and C5 conversion highlights the interconnectedness of complement proteins in these pathways ensuring a robust immune response against pathogens.
C5a has been implicated in a variety of inflammatory and autoimmune conditions. Notably C5a is associated with rheumatoid arthritis and systemic lupus erythematosus due to its role in promoting inflammation. The dysregulation of C5a activity often exacerbates these conditions. Additionally the protein is linked with other complement components such as C4 and C6 in the pathology of these diseases suggesting a potential target for therapeutic interventions aiming to modulate complement activity.
