GluK1 also known as GluR5 is a subunit of the kainate-type ionotropic glutamate receptor. It has a mass of approximately 99 kDa. This protein is expressed in various regions of the central nervous system including the hippocampus and cerebral cortex. GluK1 operates as a ligand-gated ion channel mediating synaptic transmission by allowing the influx of sodium and to a lesser extent calcium ions upon activation by kainate.
Biological function summary
GluK1 plays a role in modulating neurotransmission and synaptic plasticity. As part of the kainate receptor complex it contributes to excitatory signaling in the brain. Other components of this complex include GluK2 GluK3 GluK4 and GluK5 subunits. This receptor complex influences neuron excitability and has a hand in shaping synaptic networks by regulating glutamate release from presynaptic terminals.
Pathways
GluK1 participates in glutamatergic neurotransmission and the regulation of synaptic strength. This protein interacts with other ionotropic glutamate receptors such as AMPA and NMDA receptors within these pathways. These interactions affect long-term potentiation (LTP) and long-term depression (LTD) which are essential for learning and memory processes in the brain.
Dysfunction or dysregulation of GluK1 has been linked to neurological disorders such as temporal lobe epilepsy and schizophrenia. Abnormal GluK1 activity can influence neural excitability which is connected to the onset and progression of epileptic episodes. Moreover changes in GluK1 function may be associated with altered glutamatergic signaling in schizophrenia alongside proteins like NMDAR highlighting its importance in neuropsychiatric conditions.
