DHHC-15 also known as ZDHHC15 is a member of the DHHC domain-containing protein family. This protein functions as a palmitoyltransferase catalyzing the addition of palmitate a 16-carbon fatty acid to specific cysteine residues in substrate proteins. It exhibits an approximate molecular mass of 37-40 kDa. Researchers have found DHHC-15 predominantly in the brain but it is not limited to this organ as it also expresses in other tissues like the heart and lungs. This suggests its diverse role in cellular activities across different tissues.
Biological function summary
DHHC family members contribute to the regulation of protein trafficking stability and function. They often function as subunits within larger protein complexes influencing signaling pathways by modifying protein-membrane associations. DHHC-15 precisely modifies proteins that are important for neurotransmission. By doing so DHHC-15 influences synaptic signaling and plasticity impacting how neurons communicate and adapt in the central nervous system. Its palmitoylation activity ensures proper localization and function of target proteins enhancing cellular communication.
Pathways
DHHC-15 influences protein palmitoylation pathways which are essential in post-translational modifications that affect protein targeting and function. The protein is intimately involved in the Ras signaling pathway which is essential for cell growth and differentiation. Proteins such as H-Ras and N-Ras undergo palmitoylation by DHHC-15 which in turn regulates their membrane association and signaling activity. These interactions highlight the importance of DHHC-15 in modulating key intracellular signaling routes and cellular responses.
Alterations in DHHC-15 have correlations with neurodegenerative diseases and intellectual disabilities. Modified DHHC-15 expression or function might contribute to the pathology of Alzheimer's disease by affecting the processing or localization of Alzheimer-related proteins. Additionally mutations or dysfunctions in DHHC-15 have associations with intellectual disability disorders possibly linked to its role in modifying proteins important for brain development and function. Its interactions with APP (amyloid precursor protein) further underline its significance in disease pathology providing a potential therapeutic target in these conditions.
