GRAMD1B also called Aster-B is a phospholipid transporter with a mass of approximately 89 kDa. It plays a significant role in cholesterol regulation by transferring cholesterol from the plasma membrane to the endoplasmic reticulum. GRAMD1B is expressed in various tissues including the liver and brain. Its activity depends on the binding to phosphatidylserine-rich domains within the plasma membrane. This protein also contains a GRAM domain which aids in its lipid-binding properties and interaction with membranes enriched in specific lipids.
Biological function summary
This protein assists in maintaining lipid homeostasis by facilitating cholesterol transport. GRAMD1B performs its function as part of a larger protein complex that includes GRAMD1A and GRAMD1C. This association suggests a cooperative mechanism in regulating lipid distribution and dynamics within the cell. Through this complex GRAMD1B contributes to membrane biogenesis and possibly plays a role in cellular signaling pathways that rely on cholesterol and specific phospholipids.
Pathways
GRAMD1B is involved in the phosphoinositide signaling pathway where its lipid-binding properties are essential. The protein interacts with other important signaling molecules like AKT and PI3K influencing downstream signaling cascades that affect cellular growth and metabolism. Additionally GRAMD1B might play a role in the sphingolipid metabolism pathway where its actions could alter the composition of lipid microdomains within the cell membrane and affect signal transduction.
GRAMD1B has been linked to metabolic disorders such as non-alcoholic fatty liver disease (NAFLD). Research suggests that dysregulation of cholesterol transport could contribute to the development of NAFLD. Furthermore GRAMD1B shows a connection to neurodegenerative diseases through interactions with proteins like ApoE which plays a role in lipid transport and is associated with Alzheimer's disease. This connection suggests that alterations in lipid homeostasis due to GRAMD1B dysfunction may contribute to the pathophysiology of these disorders.
