ProCNP also known as pro-C-type natriuretic peptide is an inactive precursor to C-type natriuretic peptide (CNP) a member of the natriuretic peptide family. ProCNP has a molecular mass of approximately 12 kDa. This peptide is particularly expressed in the central nervous system bones and blood vessels. CNP is widely distributed in brain tissues and offers significant biological effects in these areas which are contrasted to the limited distribution and distinct functions of the other natriuretic peptides.
Biological function summary
Pro-C-type natriuretic peptide plays an important role in the regulation of vascular tone and bone growth. It does not act on its own but must be processed into active CNP to exert its functions. CNP does not work as part of a complex but directly interacts with particulate guanylyl cyclase membrane receptor-B (NPR-B) which mediates its effects on target tissues by increasing cyclic guanosine monophosphate (cGMP) levels.
Pathways
ProCNP influences two main pathways: the natriuretic peptide signaling pathway and bone development pathways. In particular CNP derived from proCNP regulates endochondral ossification and promotes chondrocyte differentiation. This regulation is essential in growth plate development and bone lengthening. CNP associated with natriuretic peptide receptor B contrasts with atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) which more intensely modulate cardiovascular functions.
ProCNP when processed to CNP demonstrates a link to achondroplasia a genetic disorder of bone growth leading to dwarfism. Alterations in the CNP-NPR-B pathway due to genetic mutations can impair normal bone development. Moreover it has an association with cardiovascular conditions where abnormal levels affect vascular homeostasis. These effects relate to the interaction with other natriuretic peptides like BNP as studies frequently measure proCNP levels alongside BNP in heart-related assessments.
