DOK2 also known as Docking Protein 2 or p56dok-2 is an adaptor protein with a molecular weight of approximately 56 kDa. It is expressed in hematopoietic cells and is found in various tissues including the spleen thymus and bone marrow. DOK2 acts mechanically by mediating signal transduction processes and interacting with several receptors particularly those involved in immune response.
Biological function summary
Within the cellular environment DOK2 plays an important role in the negative regulation of cell signaling pathways. It acts as a part of a signaling complex where it inhibits the Ras pathway by interacting with Ras GTPase-activating proteins. This interaction prevents excessive cell proliferation and contributes to maintaining cellular homeostasis. DOK2's inhibitory functions are essential in controlling cell cycle and apoptosis processes.
Pathways
DOK2 operates as a negative regulator in key signaling pathways such as the epidermal growth factor receptor (EGFR) pathway and the T cell receptor (TCR) signaling pathway. In the TCR pathway it associates with proteins like SHIP and SHC thereby playing a part in modulating T cell activation and signal progression. In these pathways DOK2’s presence prevents overactivation that could otherwise lead to pathological conditions.
Aberrant DOK2 function has been linked to the development of certain types of leukemia such as chronic myeloid leukemia (CML) where it may play a role in leukemic cell survival. Altered DOK2 expression also shows connection to autoimmune diseases potentially through dysregulation of immune cell signaling. An interaction with proteins like BCR-ABL in CML illustrates DOK2’s involvement in disease states making it an important focus for research in targeted therapies.
