Product details:
Abcam's 6-keto-PGF
in vitro
competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of 6-keto-PGF
alpha in serum, urine, tissue culture media and other biological fluids.
A donkey anti-sheep IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-6-keto-PGF
alpha antigen and a polyclonal sheep antibody specific to 6-keto-PGF
alpha. After incubation the excess reagents are washed away. pNpp substrate is added. After a short incubation the enzyme reaction is stopped and the yellow color generated by alkaline phosphatase is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of 6-keto-PGF
captured in the plate.
6-keto-PGF
is a stable hydrolysis product of prostacyclin (PGI
). Due to the short half-life of PGI
in buffer (2-3 minutes) the production of prostacyclin is typically monitored by measurement of the 6-keto-PGF
metabolite and its derivative in urine, 2,3-dinor-6-keto-PGF
has autocoid functions that include platelet aggregation, inhibition of vasoconstrictor responses to pressor hormones, platelet anti- and disaggregatory properties, and reproductive functions. The effects are induced by receptor-mediated elevation of cAMP. Prostacyclin has clinical applications in atherosclerosis and protection from myocardial ischemia.

6-keto-PGF1α also known as 6-keto-prostaglandin F1α or simply PGF1α is a stable metabolite of prostacyclin (PGI2). Prostacyclin and its metabolites have a molecular mass of around 375 daltons. This metabolite predominantly arises from endothelial cells where it serves as an indicator of prostacyclin production. The 6-keto-PGF1α is produced following the spontaneous hydrolysis of PGI2 which is notoriously unstable in biological systems.
Biological function summary
This metabolite acts as an important marker in various physiological and pathological processes rather than having a direct biological activity. It forms part of a larger complex of prostanoids that includes other derivatives of arachidonic acid. As a breakdown product 6-keto-PGF1α does not directly engage in extensive signaling but reflects the prior synthesis and action of prostacyclin. Monitoring levels of this metabolite can provide valuable insights into the regulation and disturbances of prostacyclin pathways.
Pathways
Prostacyclin and its stable metabolite 6-keto-PGF1α play roles in the cardiovascular system by signaling within the arachidonic acid cascade. This pathway includes enzymes such as cyclooxygenase 1 and 2 (COX-1 and COX-2) and further conversion processes leading to various prostanoids. These pathways integrate with others involved in the regulation of blood flow inflammation and thrombosis with proteins like thromboxane A2 acting in opposition to prostacyclin balancing vasodilation and platelet aggregation.
6-keto-PGF1α levels are studied in conditions related to cardiovascular health such as hypertension and atherosclerosis. These conditions involve endothelial dysfunction where reduced production of prostacyclin compromises vascular homeostasis. This metabolite also relates to inflammatory disorders as disturbed prostaglandin production influences inflammation and blood vessel function. In both cases its presence pairs with other key metabolites like thromboxane A2 that offer insight into the disease mechanisms and potential therapeutic targets.
