KAT4 also known as TBP Associated Factor 1 (TAF1) is a protein with a mass of around 240 kDa. It functions mechanically as part of the transcription machinery where it acts as an important component of the transcription factor IID (TFIID) complex. This protein is expressed in various tissues with a significant expression in the liver. TAF1 is involved in the regulation of gene expression acting as a bridge between specific DNA sequences and the general transcription factors that promote RNA polymerase II activity.
Biological function summary
TAF1 is an integral part of the TFIID complex which consists of multiple subunits. It plays a critical role in the initiation of gene transcription by facilitating the binding of the TFIID complex to RNA polymerase II and other associated factors. TAF1 contributes to the accurate transcription start site selection and the regulation of promoter activity influencing gene expression across diverse cell types.
Pathways
The role of TAF1 fits into the transcription regulation and RNA polymerase II-mediated pathways. It collaborates with proteins like TBP (TATA-binding protein) to initiate transcription. TAF1's kinase activity also involves phosphorylation processes that are part of transcription regulation mechanisms connecting it to broader signaling pathways within the cell.
TAF1 has associations with neurodevelopmental and degenerative disorders such as Parkinson's disease. Mutations or dysregulation in TAF1 can lead to abnormal transcriptional activity which impacts neuronal development and survival. The protein's interaction with other transcription-related proteins including TBP provides further insight into how transcriptional misregulation can contribute to these diseases.
