AGK also known as acylglycerol kinase is an important enzyme involved in lipid metabolism. It is responsible for phosphorylating mono- and diacylglycerol to produce phosphatidic acid. The molecular mass of AGK is approximately 45 kDa. This enzyme is expressed in various human tissues with notable expression in tissues like the heart skeletal muscle and pancreas. The presence of AGK in these tissues indicates its significant role in energy metabolism and lipid regulation.
Biological function summary
AGK participates in the biosynthesis of phospholipids important for maintaining cell membrane integrity. It is also part of the TIM22 complex in the mitochondrial inner membrane which is important for the import and assembly of proteins. This role shows that AGK not only influences lipid metabolism but also affects mitochondrial function and structure. Its activity is critical in tissues that require high energy or have a high turnover of membrane lipids.
Pathways
AGK plays a role in both the glycerolipid and phosphatidylinositol signaling pathways. In the glycerolipid pathway AGK acts to regulate the levels of lipid intermediates which are essential for diverse cellular processes. In the phosphatidylinositol pathway AGK indirectly influences processes like cell growth and survival interacting with other proteins such as PI3K and mTOR. This connects AGK to a wider network of signals impacting cellular functions and health.
AGK is linked to mitochondrial disorders particularly Sengers syndrome which affects energy metabolism. Mutations in AGK can lead to problems in the mitochondrial function highlighting its significance in mitochondrial diseases. Furthermore AGK is associated with some cancers as its dysregulation can influence tumor progression by altering lipid signaling. The interaction of AGK with proteins such as BRAF in cancer shows how it can alter cellular proliferation and survival pathways impacting disease development and progression.
