The '5HT1D Receptor' also known as the serotonin receptor 1D is a protein that plays an important role in neurotransmission. It is a G-protein coupled receptor (GPCR) with a molecular mass of approximately 47 kDa. This receptor is widely expressed in the human central nervous system particularly in the brain regions such as the basal ganglia and the frontal cortex. The 5HT1D receptor is involved in inhibiting the release of serotonin and other neurotransmitters acting as an auto-receptor in the serotonergic pathways.
Biological function summary
The 5HT1D receptor functions in the regulation of mood anxiety and vasoconstriction. It does not form part of a known receptor complex but interacts with intracellular signaling molecules to produce its effects. The receptor modulates the release of various neurotransmitters including dopamine and norepinephrine thereby influencing neurological processes and behaviors. Its role is critical in managing the serotonergic tone within different central nervous system components.
Pathways
The 5HT1D receptor is integrated into the serotonergic signaling pathway impacting both neurological and peripheral systems. It interacts with other serotonin receptors such as 5HT1B which also participate in controlling neurotransmitter release. The cAMP signaling pathway is another significant pathway where the receptor plays a part by inhibiting the production of cAMP ultimately affecting neuronal excitability and response.
The 5HT1D receptor is notably associated with migraines and depression. The receptor’s function in regulating serotonin release makes it a target for migraine treatments as alterations in serotonin levels are linked with migraine pathophysiology. In the context of depression 5HT1D receptors are studied for their potential role in the disorder's symptomatology and treatment often in connection with other proteins such as serotonin transporters (SERT) that influence serotonin availability in synaptic clefts.
