CD13 also known as aminopeptidase N is a zinc-dependent enzyme with a mass of approximately 150 kDa. It features on the surface of various cells especially in epithelial and endothelial cells as well as on fibroblasts. CD13 is recognized for its role in the hydrolysis of peptides by removing N-terminal amino acids thereby regulating diverse physiological processes. The expression of CD13 is widespread being significant in tissues like the kidney intestine and placenta. Bestatin also known as ubenimex serves as a specific inhibitor of aminopeptidase demonstrating the key position CD13 holds in peptide modulation.
Biological function summary
Aminopeptidase N impacts immune system processes including antigen presentation and inflammatory responses. CD13 forms interactions with components of the extracellular matrix and influences cell signaling pathways that modulate immune cell adhesion and migration. Although not necessarily part of a larger stable complex its enzymatic activity ubiquitously affects peptide turnover and cell surface antigen modification. CD13’s broader roles extend to angiogenesis and cellular differentiation underlining its multi-functional capacity in biological systems.
Pathways
CD13 actively contributes to processes in the renin-angiotensin system and the leukotriene pathway. In the renin-angiotensin system it participates in regulating blood pressure and fluid balance interacting with other enzymes such as ACE to modulate angiotensin levels. Within the leukotriene pathway CD13 facilitates the conversion of leukotriene A4 into its subsequent forms working alongside leukotriene A4 hydrolase. This connection highlights its influence in inflammatory responses and further integrates it with key metabolic and signaling pathways maintaining physiological homeostasis.
Aminopeptidase N holds significant linkage with cancer and inflammatory diseases. Its altered expression or activity links to tumor progression and metastasis emphasizing the enzyme's influence on tumor cell proliferation and invasion. Additionally CD13 associates with chronic inflammatory conditions like rheumatoid arthritis where it modulates inflammatory cell migration and function. Interacting proteins such as those in leukotriene regulation can modify disease outcomes positioning CD13 as both a potential biomarker and therapeutic target in these pathological contexts.
