Ikaros also known as IKZF1 is a zinc finger transcription factor with a mass of around 58 kDa. It is important for the development and function of the immune system specifically impacting lymphocyte differentiation. Ikaros is prominently expressed in hematopoietic cells including both B and T lymphocytes. IKZF3 another member of the Ikaros family serves similar roles while cereblon (CRBN) is part of the E3 ubiquitin ligase complex. CRBN is not a transcription factor but functions in protein ubiquitination and is expressed in multiple tissues suggesting diverse functions across different biological contexts.
Biological function summary
Ikaros and IKZF3 act as transcriptional regulators shaping lymphocyte gene expression in the immune system. They form complexes with other proteins to exert their regulatory effects on chromatin and transcription. CRBN as a substrate receptor for the CUL4-RBX1-DDB1-CRBN E3 ubiquitin ligase complex influences protein degradation pathways. It does not directly regulate transcription but plays a part in controlling protein turnover which can have regulatory impacts on cell signaling pathways.
Pathways
The significance of Ikaros and IKZF3 extends to the JAK-STAT signaling and B-cell receptor signaling pathways. In these pathways their transcriptional regulation affects gene expression critical for immune cell function. CRBN's role through ubiquitination connects indirectly to these pathways by modulating protein stability. Together they interact within complex networks of protein regulation influencing cellular responses to external stimuli.
Ikaros and IKZF3 are linked to hematologic malignancies such as leukemia. These transcription factors' mutations or dysregulation can lead to uncontrolled cell proliferation. The connection of CRBN to diseases like multiple myeloma comes from its interaction with the drug lenalidomide a derivative of thalidomide which modulates the immune response by degrading IKZF proteins. These interactions highlight the therapeutic targeting potential of CRBN in relation to drug efficacy and disease modulation.
