Gli3 also known as GLI-Kruppel family member 3 is a transcription factor part of the Gli protein family. It weighs approximately 170 kDa. Gli3 is expressed in various tissues including the brain lungs and limbs. It modulates gene expression important for organ development and differentiation. Gli3 can function as both a repressor and an activator of transcription depending on its processing into a shorter repressor form or remaining a full-length activator form.

It does not usually act alone; instead it is part of the Hedgehog signaling pathway complex which includes related proteins like Gli1 and Gli2.
