FKBP15 also known as FK506-binding protein 15 is a protein that functions as a peptidyl-prolyl cis-trans isomerase facilitating protein folding. It possesses a molecular mass of approximately 20 kDa. This protein is expressed mainly in tissues such as the brain and testis but can also be found in other tissues at lower levels. The primary role of FKBP15 involves protein folding and stabilization which is important for maintaining cellular functions.
Biological function summary
FK506-binding protein 15 assists in modulating protein-protein interactions due to its isomerase activity. FKBP15 is involved in the secretory pathway and may associate with various intracellular complexes to exert its function. This association enhances its ability to regulate proteins trafficking and processing within the endoplasmic reticulum ensuring proteins reach their correct destinations within the cell.
Pathways
FKBP15 plays a role in intracellular signaling and immune regulation. It participates in the calcineurin signaling pathway where it regulates calcium ion-dependent processes linking it to proteins such as calcineurin and NF-AT. FKBP15 also engages with the endoplasmic reticulum-associated degradation (ERAD) pathway contributing to the degradation of misfolded proteins and maintaining cellular homeostasis.
FKBP15 has links to neurodegenerative disorders and cancer. In neurodegeneration disrupted FKBP15 function can affect protein folding balance connecting to proteins like tau which is implicated in Alzheimer's disease. In cancer FKBP15's role in protein folding and signaling pathways may influence cellular proliferation and tumor development providing a potential therapeutic target for drug development.
