SEI-1 also known as p34SEI-1 or CDK4-binding protein p34 is an important protein involved in cell cycle regulation. It has a molecular mass of approximately 31 kDa. Expression of SEI-1 occurs in various tissues with a noticeable presence in proliferative tissues like the thymus spleen and certain cancerous tissues. It exerts its role mechanistically by interacting with cyclin-dependent kinases (CDKs) particularly CDK4 and modulating their activity during cell cycle progression.
Biological function summary
SEI-1 influences cell cycle regulation and proliferation. It acts as a regulatory factor for the CDK4/Cyclin D1 complex facilitating the G1 to S phase transition in the cell cycle. This protein by interacting directly with CDK4 plays a part in the assembly and activation of CDK4 complexes which are pivotal for proper cell division. SEI-1 also has a role outside the cell cycle like apoptosis regulation hinting at diverse functionality within cellular processes.
Pathways
SEI-1 sits prominently in the cell cycle control pathway. It associates closely with the retinoblastoma protein (Rb) pathway where its modulation of CDK4 activity influences Rb phosphorylation. SEI-1’s interaction with CDK4 links it to both the MAP kinase (MAPK) and PI3K/Akt signaling pathways impacting cell growth and survival. Through these associations SEI-1 contributes importantly to the regulation of cellular proliferation and survival mechanisms.
SEI-1 shows connections to cancer particularly in its overexpression found in several cancer types such as breast cancer and gliomas. Its ability to amplify CDK4 activity and deregulate the cell cycle links SEI-1 with oncogenesis. Therapeutic interest targets its interaction with CDK4 to control cancer cell proliferation. SEI-1 also associates with diabetes as its pathway involvement connects to insulin signaling potentially influencing insulin resistance.
